Purpose: Vascular targeted photodynamic therapy (VTP) is a nonsurgical tumor ablation approach used to treat early-stage prostate cancer and may also be effective for upper tract urothelial cancer (UTUC) based on preclinical data. Toward increasing response rates to VTP, we evaluated its efficacy in combination with concurrent PD-1 inhibitor/OX40 agonist immunotherapy in a urothelial tumor-bearing model. Experimental design: In mice allografted with MB-49 UTUC cells, we compared the effects of combined VTP with PD-1 inhibitor/OX40 agonist with those of the component treatments on tumor growth, survival, lung metastasis, and antitumor immune responses. Results: The combination of VTP with both PD-1 inhibitor and OX40 agonist inhibited tumor growth and prolonged survival to a greater degree than VTP with either immunotherapeutic individually. These effects result from increased tumor infiltration and intratumoral proliferation of cytotoxic and helper T cells, depletion of Treg cells, and suppression of myeloid-derived suppressor cells. Conclusions: Our findings suggest that VTP synergizes with PD-1 blockade and OX40 agonist to promote strong antitumor immune responses, yielding therapeutic efficacy in an animal model of urothelial cancer.  相似文献

A classical Lewis adduct derives from a covalent bond between a Lewis acid and a base. When the adduct formation is precluded by means of steric hindrance the association of the respective acid-base molecular system is defined as a frustrated Lewis pair (FLP). In this work, the archetypal FLP Mes3P/B(C6F5)3 was characterized for the first time by resonance Raman spectroscopy, and the results were supported by density functional theory (DFT) calculations. The charge transfer nature of the lowest energy electronic transition, from phosphine to borane, was confirmed by the selective enhancement of the Raman bands associated to the FLP chromophore at resonance condition. Herein, we demonstrate the use of resonance Raman spectroscopy as a distinguished technique to probe the weak interaction involved in FLP chemistry.  相似文献

We show an accurate procedure to obtain a Fourier transform (FT) with no dc term using a commercial twisted-nematic liquid-crystal display. We focus on the application to holographic storage of binary data pages, where a drastic decrease of the dc term in the FT is highly desirable. Two different codification schemes are considered: binary pi radians phase modulation and hybrid ternary modulation. Any deviation in the values of the amplitude and phase shift generates the appearance of a strong dc term. Experimental results confirm that the calculated configurations provide a FT with no dc term, thus showing the effectiveness of the proposal.  相似文献

Eurysternus caribaeus was analyzed cytologically by conventional chromosomal staining. The species presents a diploid number of 2n=8, chromosomes with two arms and an XY sex determining mechanism. This is the first karyotype described for the genus Eurysternus and the tribe Eurysternini, and is also the smallest diploid number observed in the family Scarabaeidae and superfamily Scarabaeoidea.  相似文献

The gas-phase behavior of 12 quinolon-4(1H)-imine derivatives with antiplasmodial activity was investigated using electrospray ionization tandem mass spectrometry together with collision induced dissociation and density functional theory (DFT) calculations. The most probable protonation site was predicted by calculating the proton affinity (PA) values for each possible protonation site and it was found to be the imine nitrogen for all compounds under study. Fragmentation pathways of the protonated molecules were proposed and the assignment of product ion structures was performed taking into account theoretical calculations. The nature of the quinoline substituent was found to influence the gas-phase behavior of the compounds under study. The data acquired allowed to bracket the proton affinity of the quinolin-4-imine scaffold, which can be a useful starting point to choose appropriate references for determining PA values of this scaffold. When starch is incorporated into puddings, desserts, and other foods containing sugar as the main ingredient, it will have an effect on the gelatinisation temperature and pasting properties. Many studies have been undertaken to investigate the effect of several sugars in foods and starches, as well as their physicochemical and functional properties. These studies have verified the significant influence on these properties, which are dependent on the nature of sugar and of starch. In this study, pinhão starch was extracted in the laboratory and was added, stirring for 60 min, to solutions at 1 % of each of the following sugars: fructose, glucose, sucrose, and 0.5 % fructose and 0.5 % glucose. After this time, the slurry was filtered, washed, dried at 40 °C and kept in a desiccator over anhydrous calcium chloride until constant mass. The effects of each sugar on the surface of the pinhão starch granules were observed using the non-contact method of atomic force microscopy, whereby it was possible to verify a decrease in the average diameter and an increase in the average roughness. X-ray diffractometry made it possible to evaluate the degree of relative crystallinity, which was proportional to the roughness and inversely proportional to the gelatinisation enthalpy (ΔH), which was studied by differential scanning calorimetry.  相似文献

A series of supermolecular liquid crystals has been synthesized by combining phosphorus dendrimers of the zero, first, and fourth generations with sulfonamide derivatives, thus generating dendromesogens bearing 6, 12, and 96 mesogenic units on their surfaces. The relevant reactions could be monitored by 1H, 19F, and 31P{1H} NMR spectroscopies. The thermal and mesomorphic properties of the products have been studied by optical microscopy, differential scanning calorimetry, and X‐ray diffraction. All of the new macromolecules prepared in this work have been found to show mesomorphic properties over a wide temperature range; moreover, for all of the compounds, the columnar mesophases observed were maintained or vitrified at room temperature. On increasing the generation of these dendromesogens, mesophases appear at lower temperatures and remain stable over a wider temperature interval. In all cases, on the basis of X‐ray analysis, a cylindrical symmetry of the molecules can be proposed to promote the supramolecular columnar arrangement observed in the mesophases. In this type of model, the height of the dendrimer clearly increases with increasing dendrimer generation, whereas its cross‐ sectional area increases only slightly, probably due to compression of the highly hyperbranched structures as a consequence of their progressive steric constraints. The mesomorphic arrangement is governed by the peripheral sulfonamide units.  相似文献

The spatial distribution of chemical compounds in sugarcane bagasse is an important issue in its use as a raw material for second generation ethanol production from cellulose hydrolysis. Lignocellulosic materials including whole bagasse, fiber, pith, and respective samples obtained after chemical bleaching were investigated using confocal fluorescence microscopy and spectroscopy with one and two-photon excitation. Autofluorescence from unbleached samples revealed that emission from fiber walls containing the lignin fraction was longitudinally oriented. After bleaching treatment, the oriented emission was partially disrupted. Autofluorescence from bleached samples with a residual lignin content of about 1 % was ascribed to improved dispersion of remaining fluorophores throughout the samples inducing a concomitant reduction of fluorescence self-quenching in the samples. The combination of autofluorescence images with spectral emission and lifetime measurements provides a tool for microscopic characterization of natural bagasse samples. Moreover, the technique allows monitoring bleaching processes related to lignin removal.  相似文献
